采用 DNA 杂交的方法,对异育银鲫及其人工杂合种进行了外源 DNA 的检测分析,发现两个雌核发育系的异育银鲫及其人工杂合种的 DNA 与红鲤 DNA 片段间的分子杂交均有阳性斑点出现,这表明,在异育银鲫及其人工杂合种产生过程中,父本(红鲤)的 DNA 片段的确可以随机地掺入到母本细胞的 DNA 中,从而产生了异精效应,使其子代生长加快,并出现父本性状。  相似文献

摘要 目的:探讨不同国际妇产科学联合会(FIGO)分期子宫内膜癌患者外周血红细胞分布宽度(RDW)、血清对氧磷酶-1(PON-1)表达水平差异及其诊断价值。方法:选取我院2018年1月到2020年1月收治的80例子宫内膜癌患者作为研究对象,根据FIGO分期对所有患者进行分组,分为Ⅰ期组23例,Ⅱ期组22例,Ⅲ期组18例,Ⅳ期组17例。另选取同期来我院体检的30名健康志愿者作为对照组,对比所有受检者RDW、PON-1表达水平,并应用ROC曲线分析外周血红细胞分布宽度、血清对氧磷酶-1表达水平对不同FIGO分期子宫内膜癌的诊断价值。对80例子宫内膜癌患者进行2年随访,将2年内死亡的25例患者分为死亡组,将其余55例患者分为存活组,对比死亡组与存活组临床一般情况与RDW、PON-1表达水平,并应用logistic回归分析分析RDW、PON-1对子宫内膜癌的预后预测价值。结果:五组受检者RDW、PON-1水平对比差异显著,Ⅳ期组RDW水平高于Ⅲ期组、Ⅱ期组、Ⅰ期组和对照组,Ⅳ期组PON-1水平低于Ⅲ期组、Ⅱ期组、Ⅰ期组和对照组(P<0.05);RDW结合PON-1联合诊断的准确度、敏感度、阳性预测值高于RDW单一诊断和PON-1单一诊断(P<0.05)。RDW诊断子宫内膜癌不同分期价值曲线下面积为89.63,最佳诊断着色界限值为75.73 %,PON-1的曲线下面积为78.89,最佳诊断着色界限值为82.53 %,两者联合的曲线下面积为84.26,最佳诊断着色界限值为87.57 %;存活组与死亡组患者性别、年龄、病理类型对比无明显差异(P>0.05),两组患者FIGO分期、基层浸润、组织分化程度、血清RDW与PON-1表达水平对比差异显著(P<0.05);logistic回归分析结果表明:基层浸润、RDW与PON-1为子宫内膜癌患者预后的独立影响因素(P<0.05)。结论:RDW、PON-1联合对子宫内膜癌FIGO分期的诊断价值较高,且基层浸润、RDW与PON-1为子宫内膜癌患者预后的独立影响因素。临床上需针对基层浸润、RDW升高与PON-1水平降低的患者采取相关措施,改善治疗方案,降低患者死亡率情况。  相似文献

Synchronously dividing cultures of the unicellular green alga Scenedesmus obtusiusculus were cultivated for 24 or 70 h in medium high (1000 μM) or low (60 μM) in phosphorus. Aliquots of AlCl3 (0, 37, 74, 111, 148, 185, or 222 μmol) were added daily to 1 l cell suspension at the end of the cell division phase. Algae were also grown in media with different pH, adjusted with HCl, in the absence of AlCl3.
Effects of Al on cell metabolism vary with the intracellular Al concentration and with the concentration of Al available per cell. When the concentration of phosphorus is low, internal concentrations of Al are high and the chlorophyll content and the net dry matter production per cell increase, whereas the photosynthesis and the cell division are increased. Presence of Al in a low P medium decreases the pH of the medium down to 4.5. There are only small effects of Al in the presence of P, due to precipitation of most of the Al with P in the medium.
Despite the Al-induced decrease of the pH of the culture medium, effects caused by Al cannot be explained as a pH effect. Instead, the Al effect may, at least to some extent, be related to a decrease in availability of P in the metabolism, due to formation of aluminium phosphate inside the cell.  相似文献

Objective: To measure red cell flux of adipose tissue in morbidly obese patients' pannus in the upright and supine position to determine factors which would render the lower pannus susceptible to ischemic necrosis. Design: A cohort study of morbidly obese subjects without ischemic necrosis. Setting: University teaching hospital. Patients: Twenty-three consecutive morbidly obese patients referred for gastroplasty. Measurements: Red cell flux, measured as RMS voltage by a laser Doppler velocimeter. An optical fiber with a tip diameter of 250μ was inserted into the upper and lower pannus and output recorded in the upright and supine positions. Other variables recorded were age, BMI, blood pressure and serum lipids. Results: Adipose tissue red cell flux demonstrates considerable spatial and temporal heterogeneity from subject to subject and in various locations in the pannus. No differences in red cell flux were detected in response to change in position. However, regression analysis demonstrated that the gradient between the upper and lower abdomen in the supine position was increasingly positive with age and in the upright position it was increasingly positive with increasing weight or BMI. Conclusions: These data suggest that red cell flux is heterogeneously distributed in the abdominal pannus and is not greatly influenced by body position. However, with increasing age and adiposity there is a gradient for decreased red cell flux to the lower portion of the pannus. This may be a factor in rendering this part of the pannus prone to ischemic fat necrosis.  相似文献

本试验研究了红穗醋栗色素的理化性质,结果表明:红穗醋栗色素热稳定性很好,而光照对其有一定的降解作用。pH值对该色素影响明显,宜在酸性食品中应用。几种金属离子Na~ 、Ca~(2 )、Al~(3 )、Zn~(2 )。Cu~(2 )对该色素的色泽没有影响;而Fe~(3 )、Sn~(2 )则有不良影响。食品中常含的几种添加物葡萄糖、蔗糖、淀粉和抗坏血酸对该色素无不良影响。红穗醋栗色素对Na_2SO_3的还原性耐性较强,对H_2O_2的耐氧化性很差。防腐剂苯甲酸钠对该色素也有一定的不良影响。  相似文献

Summary Examination was made of the structure and assembly of the cortical microtubule (MT) cytoskeleton in the coenocytic green algaBoodlea coacta (Dickie) Murray et De Toni by immunofluorescence microscopy. Cortical MTs inBoodlea protoplasts are arranged randomly but some show a meridional arrangement within 6 h after protoplast formation. At 6–9 h such MTs become highly concentrated and parallel to each other in certain areas. At 12 h the concentration is uniformly high throughout the cell, indicating the completion of high density meridional arrangement of cortical MTs. Cortical MTs exhibiting a high density, meridional arrangement show characteristic disassembly by treatment with 10 M amiprophos-methyl (APM) or cold treatment (0 °C). Disassembly occurs by each MT unit at positions skipping 30–40 m in the transverse direction, and neighboring MTs subsequently disassemble to form MT groups. Each group becomes slender and then disappears completely within the following 24 h. The meridional arrangement of cortical MTs is disrupted by N-ethylmaleimide (NEM) accompanied by a remarkable reduction in density. The remaining MTs form groups at 30–40 m intervals from each other, as also occurs with drug or cold treatment, but disruption and density return to normal levels following removal of NEM. It appears that there are meridionally oriented channels, anchor-rich and anchor-poor, in the plasma membrane. The channels could be distributed alternately and anchors could be deposited in a cross-linking manner with cortical MTs to form a stable cortical MT-cytoskeleton. The phytochrome family of signal-transducing photoreceptors provides plants with the capacity to perceive variations in the relative fluxes of red (R) and far-red (FR) radiation. This capacity has been proposed to be of ecological value in the perception of the proximity of neighbouring plants and the consequent induction of shade avoidance responses. The work reported here has evaluated this potential by determining quantitatively the effect of neighbour proximity on the growth of canopies of Populus trichocarpa×deltoides‘Beaupré’ trees, and relating the measured variables to the long-term vectoral radiation quality inside each canopy. The spectral distribution of radiation inside four canopies of Populus trichocarpa×deltoides‘Beaupre’ of different densities was monitored throughout the growing season. Spectral distributions inside the canopies were measured in 10° wedges at different heights and angles. The results are presented as PFD over 400–700 nm (PFD400–700) and PFD over 400–800 nm (PFD400–700). Results are also presented for the calculated phytochrome photoequilibrium (Pfr/P) and red:far-red ratio (R:FR). Data are presented as in-canopy angular and height profiles, and as diurnal and seasonal variations. PFD400–700 and Pfr/P were found to be reduced inside each canopy, the reduction being greatest in the most dense canopy, and least in the most open canopy. At any height within each canopy, calculated Pfr/P decreased linearly with time throughout the growing season, until leaf senescence began. The reduction was greater in the denser canopies and was found to be similar for three consecutive field seasons. Linear relationships were found between plant stem growth rate, plant spacing and Pfr/P calculated from radiation propagated approximately horizontally within the canopies. The findings support the role of phytochrome in proximity perception in the natural environment and provide a quantitative basis for investigating the competitive interactions between plants growing in dense stands. The hypothesis is proposed that the dynamics of developing or regenerating canopies can be accounted for on the basis of phytochrome-mediated perception of the proximity of neighbouring plants.  相似文献

Aphelenchoides resinosi n. sp. and Ektaphelenchus joyceae n. sp. are described and illustrated from red pines of the Allegheny plateau of Maryland, USA. The new species were found in trees infested with Bursaphelenchus xylophilus. Primary diagnostic characters of A. resinosi females are constriction of the head, basal stylet knobs, tong postuterine sac, two incisures in the lateral field, and conical tail four to five anal body widths long with a simple terminal mucro. Diagnostic characters of the males are two pairs of subventral caudal papillae and spicule shape: Primary diagnostic characters of E. joyceae females are a slight constriction of the head, six similar lips, conical tail, and short postuterine sac. Diagnostic characters of the males are spicule size and shape, a single row of spermatocytes, and one pair of caudal papillae. Within-tree distributions of A. resinosi and E. joyceae are presented. A total of 70% of both red-needled and chlorotic-needled trees in the study were positive for A. resinosi and E. joyceae. Branch hierarchy was related to the percentage of samples positive for A. resinosi.  相似文献

Human and murine blood cells treated with ZnCl2 and bis(sulfosuccinimidyl)suberate (BS3) (a cross linking agent) undergo band 3 clustering and binding of hemoglobin to red blood cell membrane proteins. These clusters induce autologous IgG binding and complement fixation, thus favouring the phagocytosis of ZnCl2/BS3 treated cells by macrophages. The extension of red blood cell opsonization can be easily modulated by changing the ZnCl2 concentration in the 0.1–1.0 mM range thus providing an effective way to affect blood cell recognition by macrophages. In fact, murine erythrocytes treated with increasing ZnCl2 concentrations have proportionally reduced survivals when reinjected into the animal. Furthermore, the organ sequestration of ZnCl2/BS3 treated cells strongly resembles the typical distribution of the senescent cells. Since the ZnCl2/BS3 treatment can also be performed on red blood cells loaded with drugs or other substances, this procedure is an effective drug-targeting system to be used for the delivery of molecules to peritoneal, liver and spleen macrophages.  相似文献

The effects of experimental factors on protoplast formation of Prototheca zopfii Kru¨ger in 0.85 m NaCl using Macerozyme R-200 were studied based on a fractional factorial experimental design. The rate of protoplast formation was mainly affected by the incubation temperature and the age of algal cells. The optimal condition for the maximum protoplast yield was determined based on a response surface model. These were: mid-logarithmic phase cells and Macerozyme concentration of 4% at a temperature of 35°C.  相似文献
